from the group of polyphenols, alkaloids and glycosides on healthy and cancer human model cell systems with the perspective of identifying intracellular mechanisms and their potential use for improving consumer health as well as for the prevention of civilization diseases.<\/p><p>The scientific focus of the proposed project is divided into three stages:<\/p><p><strong>Stage 1<\/strong>\u00a0(Slovak side) will be focused on\u00a0<em>in vitro<\/em>\u00a0testing of selected phytonutrients from the group of polyphenols, alkaloids and glycosides (punicalagin, elagic acid, quercetin, resveratrol, apigenin, capsaicin, papaverine, berberine and others) on human model cell lines (ovarian granulosa cell line HGL5, ovarian carcinoma line, etc.) with the aim of to identify intracellular mechanisms, through which the relevant plant bioregulators act (cell viability, secretory activity, proliferation and apoptosis at the genomic, transcriptomic and proteomic level).<\/p><p><strong>Stage 2<\/strong>\u00a0(Czech side) will be focused on\u00a0<em>in vitro<\/em>\u00a0testing of selected phytonutrients from the group of polyphenols, alkaloids and glycosides (punicalagin, elagic acid, quercetin, resveratrol, apigenin, capsaicin, papaverine, berberine and others) on human dendritic cells developed from CD14+ blood mononuclear cells and on human blood BDCA-1+ (CD1c+) dendritic cells. We will analyze cell viability and apoptosis of mentioned cells. Moreover, we will test effects of plant origin bioregulators on gene expression of model cell lines (human femoral osteoblast) related to quantification of bone matrix, membrane transport and cellular signaling pathway proteins.<\/p><p><strong>Stage 3<\/strong>\u00a0(French side) will be focused on evaluating the digestion and absorption processes of phytonutrients before reaching the targeted cells. We will then be able to collect, purify and extract the nutrient metabolites for testing them on targeted cells (such as ovarian cells, mononuclear and dendritic
